Product Specific Information

The HM48-1 monoclonal antibody reacts with mouse CD48 antigen (also known as BCM1, Blast-1, and OX-45). CD48 is a 45 kDa GPI-linked glycoprotein expressed on most hematopoietic cells and plays a role in adhesion and T cell activation. It interacts with CD2 and CD244 and is useful for identifying hematopoietic progenitor subsets.

This antibody can block CD48/CD2 and CD48/CD244 interactions, inhibit spleen cell proliferation, and modulate immune responses. Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume).
Cell number range: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample.

Strain-specific Note:
NovaFluor Yellow 700 shows non-specific B-cell staining in SJL mice but not in BALB/c or C57BL/6 strains.

Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily. It mediates cell adhesion and signaling among hematopoietic cells, serving as a ligand for CD2 and 2B4. CD48 expression is elevated in certain infectious and allergic conditions and is useful as a marker for GPI-anchor defects such as paroxysmal nocturnal hemoglobinuria (PNH).
