Target Information

The expression of DUSP1 gene is induced in human skin fibroblasts by oxidative or heat stress and growth factors. It encodes a protein similar to non-receptor-type protein-tyrosine phosphatases and has significant amino acid sequence similarity to a Tyr/Ser-protein phosphatase encoded by the late gene H1 of vaccinia virus.
The bacterially expressed and purified DUSP1 protein exhibits intrinsic phosphatase activity, specifically inactivating mitogen-activated protein (MAP) kinase by dephosphorylating both phosphothreonine and phosphotyrosine residues.
It also suppresses MAP kinase activation by oncogenic ras in Xenopus oocyte extracts, suggesting an important role in cellular stress response and negative regulation of proliferation.
