Target Information
Thymidylate Synthase (TS) catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), a key step in DNA biosynthesis. TS is a major target of fluoropyrimidine-based antineoplastic drugs used in solid tumor treatments. Drugs such as 5-fluorouracil (5-FU) and fluorodeoxyuridine form a ternary covalent complex with TS in the presence of 5,10-methylenetetrahydrofolate, leading to enzyme inactivation. Expression of TS correlates with response to 5-FU in colorectal, gastric, head and neck, and breast carcinomas.
