Product Specific Information

Description:
The 93 monoclonal antibody reacts with an epitope shared by mouse CD16 and CD32.
CD16 (Fc gamma III Receptor) and CD32 (Fc gamma II Receptor) are low-affinity receptors for the mouse IgG Fc portion and are expressed by B cells, monocyte/macrophages, NK cells, and neutrophils.

Applications Reported:
This antibody has been reported for use in flow cytometric analysis.

Applications Tested:
Tested on mouse splenocytes; can be used at ≤0.5 µg per test (100 µL final volume).
Cell number may range from 10⁵–10⁸ cells/test.
Careful titration is recommended for optimal performance.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser line (405 nm) and emits at 436 nm.
Recommended filter: 450/50 bandpass or equivalent.
Ensure instrument compatibility with this fluorochrome.

Buffer Recommendation:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD16:
A 50–65 kDa cell surface molecule existing in two forms:

• Transmembranous form (NK cells, some T cells)
• Phosphatidylinositol-linked form (granulocytes)
  Acts as a low-affinity receptor for IgG (FcR III) and mediates ADCC by NK cells.

CD32:
A 40 kDa glycoprotein acting as a low-affinity receptor for IgG (Fc gamma RII).
Functions include endocytosis, secretion activation, cytotoxicity, and immunomodulation.
Expressed by B cells, monocytes, granulocytes, and platelets.
Involved in immune complex phagocytosis and B-cell antibody regulation.
Variants of CD32 may increase susceptibility to systemic lupus erythematosus (SLE).
Several transcript variants encoding different isoforms have been identified.
