Product Specific Information

The 61D3 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, macrophages, and some dendritic cells. CD14 binds LPS-LBP complexes with high affinity on monocyte surfaces.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells
• Recommended Use: 5 µL (0.5 µg) per test in 100 µL final volume (10⁵–10⁸ cells/test)
• Fluorochrome Details: Super Bright 436 excited at 405 nm, emits at 436 nm; use a 450/50 bandpass filter or equivalent
• Buffer Recommendation: Use Super Bright Complete Staining Buffer (Product # SB-4401) when multiple Super Bright dyes are used to minimize polymer interactions

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 participates in immune responses, apoptotic cell binding, and inflammatory processes.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and Croup.
