Target Information

The leptin (OB-R) receptor is a member of the cytokine receptor superfamily that plays a key role in mammalian body weight homeostasis and energy balance. Activation by leptin triggers downstream signaling through the JAK/STAT pathway. OB-R forms homo-dimers to initiate intracellular signaling. Multiple isoforms with differing signaling capabilities exist. The OB-R long form is expressed in the choroid plexus and hypothalamus, regions associated with satiety and energy regulation. Mutations in OB-R (db) or leptin (ob) genes lead to early-onset obesity and metabolic abnormalities such as insulin resistance, hyperglycemia, and altered CNS activity.
