Target Information

Voltage-gated potassium (Kv) channels are essential components of potassium currents in the heart and central nervous system.

• In the heart, Kv4 channels are involved in the repolarization phase of the action potential.
• In the brain, these channels prevent reverse-propagation of action potentials.

Associated with Kv4 channels are calcium-binding proteins termed KChIPs (Kv channel interacting proteins).
KChIPs bind to the cytoplasmic amino termini of Kv4 alpha-subunits and modulate their function.

There are three known KChIPs:

1. KChIP1 – expressed in the brain
2. KChIP2 – expressed in the brain, heart, and lung (three isoforms: a, b, c)
3. KChIP3 – also known as calsenilin or DREAM (DRE-antagonist modulator), expressed in the brain and testis

KChIP3 not only modulates Kv4 currents but also binds to regulatory DRE elements to repress transcription from the early-response gene c-fos.
It is the first known calcium-binding protein to function as a DNA-binding transcriptional regulator.
KChIP3 is also associated with presenilins and implicated in beta-amyloid formation.
