Target Information

This gene encodes a transmembrane receptor commonly referred to as DC-SIGN, expressed on dendritic cells and macrophages. It plays a key role in innate immunity by recognizing diverse pathogens including parasites and viruses.
The protein has three domains:

  1. N-terminal transmembrane domain
  2. Tandem-repeat neck domain
  3. C-type lectin carbohydrate recognition domain

The extracellular region functions as both a pathogen recognition receptor and a cell adhesion receptor. The neck domain mediates homo-oligomerization, enhancing multivalent ligand binding. Variations in the neck domain repeats affect ligand-binding affinity.
DC-SIGN is closely related to L-SIGN (GeneID 10332), with distinct ligand-binding properties and tissue distribution. Alternative splicing generates multiple isoforms.
