Target Information

CD44 is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, white matter of the brain, and certain epithelial cells of the intestine and breast. Multiple isoforms exist, with CD44H being predominant in normal tissues. It serves as a receptor for hyaluronic acid (HA) and plays roles in cell-cell interactions, adhesion, and migration. CD44 is involved in lymphocyte activation, recirculation, and homing, and its expression may increase in some carcinomas, potentially correlating with metastatic potential. It is expressed by hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia of cultured keratinocytes. Bone marrow myeloid cells and memory T cells express high levels of CD44, while peripheral B and T cells can upregulate CD44 upon stimulation. The CD44 gene undergoes complex alternative splicing, generating multiple functionally distinct isoforms. Dysregulation of CD44 is associated with diseases such as superficial keratitis and lichen sclerosus and may be related to tumor metastasis.
