• For immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD44 cell surface antigen is a 100 kDa type I transmembrane glycoprotein widely expressed on human leukocytes, white matter of the brain, and some epithelial cells of the intestine and breast. Several isoforms exist, including the predominant CD44H isoform found in many normal tissues.
CD44 acts as a receptor for hyaluronic acid (HA) and is involved in cell-cell interactions, adhesion, and migration. It participates in lymphocyte activation, recirculation, and homing. Expression may be upregulated in certain carcinomas, possibly linked to metastatic potential.
CD44 is expressed by hematopoietic and non-hematopoietic cells, epithelial tissues, and filopodia in cultured keratinocytes. Bone marrow myeloid cells and memory T cells express high levels of CD44, and peripheral B and T cells can upregulate CD44 in response to stimulation.
Alternative splicing of CD44 transcripts results in multiple isoforms with distinct functions. CD44 dysfunction is associated with diseases such as superficial keratitis and lichen sclerosus, and may be related to tumor metastasis formation.
