Product Specific Information
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and monocytes. CD4 serves as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for normal human peripheral blood cells at 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
NovaFluor dye compatibility:
Not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):
Required for optimal staining with NovaFluor conjugates. Add 5 µL per sample (regardless of number of NovaFluors). For single-color controls, use 5 µL per 100 µL cell sample.

NovaFluor conjugates are based on Phiton™ technology using engineered nucleic acid dye structures for optimized fluorescent signatures with reduced spillover and spread.

Target Information
CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. It is primarily expressed in T helper cells, monocytes, macrophages, and dendritic cells. Tissue expression includes thymus, lymph nodes, tonsils, spleen, and specific regions of the brain and gut. CD4 functions in T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
