Product Specific Information

Description:
The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, a subset of mature T helper cells, and at low levels on monocytes. CD4 serves as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reaction. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Validated for flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (0.1 µg) per test in 100 µL final volume.
Cell number: 10⁵–10⁸ cells/test (empirically determined).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails prior to combining with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology based on nucleic acid dye structures, engineered for optimized fluorescence signatures with minimal spillover and spread.

Target Information

CD4 is a co-receptor for MHC class II molecules and HIV. It is primarily expressed on T helper cells, monocytes, macrophages, and dendritic cells. At the tissue level, CD4 is found in thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. It plays a key role in T-cell activation via association with the T-cell receptor complex and Lck kinase, and may mediate neuronal damage in immune-related CNS diseases.
