Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed by most thymocytes, mature T helper cells, and at low levels on monocytes. CD4 acts as a receptor for HIV, and SK3 blocks HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells.
Recommended usage: 5 µL (0.1 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample for antibody cocktails or single-color controls.

NovaFluor conjugates utilize Phiton technology with nucleic acid dye structures engineered for optimized fluorescence and minimal spectral spillover.

Target Information

CD4 antigen recognizes MHC class II molecules and serves as a co-receptor for HIV. It is primarily expressed in T helper lymphocytes and also found in monocytes, macrophages, and dendritic cells. CD4 expression is detectable in thymus, lymph nodes, tonsils, spleen, and certain non-lymphoid tissues. It plays a role in T-cell activation and may mediate neuronal damage in immune-related CNS diseases. Multiple alternatively spliced transcripts are known (RefSeq, July 2017).
