Target Information
iNOS (Inducible Nitric Oxide Synthase, NOS)는 세포 간 다양한 신호를 전달하는 무기성 기체 자유 라디칼입니다. 혈관 이완, 신경전달, 세포독성 등 다양한 생리 반응에 관여하며, NO 생산은 NOS 패밀리 단백질에 의해 매개됩니다.
iNOS는 간에서 발현되며, lipopolysaccharide와 특정 cytokine의 조합에 의해 유도됩니다. NOS는 L-arginine을 산화시켜 L-citrulline과 NO를 생성하며, 세 iso폼(b/nNOS, eNOS, iNOS)이 알려져 있습니다.
iNOS는 macrophage, hepatocyte, synoviocyte, smooth muscle cell 등 다양한 세포에서 발견되며, IFN-γ, TNF, IL-1, IL-2, LPS 등의 자극에 의해 발현이 증가합니다. Cytokine 유도 후 iNOS는 지연된 활성 반응을 보이며 장기간 NO 생산을 지속합니다.
iNOS 관련 pseudogene은 chromosome 17의 Smith-Magenis syndrome 영역에 위치하며, iNOS 기능 이상은 achalasia 및 impotence 등과 연관됩니다.
