Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of CD22α and CD22β chains (130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas.

The extracellular domain contains seven Ig-like domains capable of binding sialic acid moieties on epithelial, endothelial, B, and T cells. The intracellular domain includes six tyrosine residues within ITIM and activation-like motifs, phosphorylated upon B-cell receptor engagement, enabling both positive and negative regulation of B-cell receptor signaling.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg)/test in a final volume of 100 µL. Cell number: 10⁵–10⁸ cells/test.

Super Bright 436 can be excited with the violet laser (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation and is limited to mature B cells and plasma cells, useful for phenotyping mature leukemias. Strongly expressed in hairy cell leukemia, diffuse large B-cell lymphoma, and nodular lymphocyte predominance Hodgkin’s lymphoma; negative in classical Hodgkin’s lymphoma.

CD22 preferentially binds α2,6-linked sialic acid. Ligand-induced tyrosine phosphorylation plays a role in regulation of B-cell receptor signaling, interacting with Src family kinases and recruiting phosphatases to modulate signal transduction.
