Product Specific Information

The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed on developing and mature B cells but not on plasma cells. Low levels may be detected on a subset of mature T cells. CD20 is implicated in B-cell activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Fluorochrome Information:
Super Bright 436 is excited by the violet laser line (405 nm) and emits at 436 nm. Recommended filter: 450/50 bandpass or equivalent. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, some T lymphocytes, and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 remains on the cell surface after antibody binding, making it a valuable target for therapeutic B-cell depletion.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
