Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Rat IFN-gamma. Anti-Rat IFN-gamma-specific antibody was purified by affinity chromatography employing an immobilized Rat IFN-gamma matrix.

Sandwich ELISA:
To detect Rat IFN-gamma by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Rat IFN-gamma (500-P122) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Rat IFN-gamma.

Western Blot:
To detect Rat IFN-gamma by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used in conjunction with compatible secondary reagents, the detection limit for Recombinant Rat IFN-gamma is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.

IFN gamma (Interferon gamma, Type II interferon)
IFN gamma is a macrophage activation factor and immune interferon produced primarily by T-lymphocytes and natural killer cells in response to antigens, mitogens, and other cytokines. It is a dimeric glycoprotein consisting of two 146 amino acid subunits (~45 kDa functional homodimer). On SDS-PAGE, IFN gamma appears as 25, 20, and minor 15.5 kDa bands due to differential glycosylation.

The biological activity of IFN gamma is highly species-specific. Human IFN gamma does not cross-react with mouse. Functions include:

• Antiviral and antitumor activity
• Induction of MHC class I and II
• Macrophage activation
• Enhanced immunoglobulin secretion by B lymphocytes

IFN gamma acts synergistically with other cytokines via IFN gamma receptor I and II, activating the JAK-STAT pathway. It shares ~40% sequence homology between human and mouse IFN gamma. Expression is upregulated by IL2, FGF basic, EGF, and downregulated by vitamin D3 or DMN. Mutations in the IFN gamma gene are associated with aplastic anemia.
