Target Information

Lysosomal enzymes containing one or two mannose 6-phosphate (man6P) moieties are transported in the cell by approximately 300 kDa cation-independent mannose 6-phosphate receptors (CI-MPR).
The biosynthetic cycle involves MPR-mediated transport of newly synthesized lysosomal enzymes from the trans-Golgi network to late endosomes or early lysosomes.
The endocytic cycle moves extracellular lysosomal enzymes from the plasma membrane to early endosomes via clathrin-coated vesicles.
MPRs cycle between these compartments every 3 hours, with most located in late endosomes, fewer in the trans-Golgi network, and about 10% at the cell surface.
MPRs also have a binding site for type II insulin-like growth factor (IGF-II), enabling simultaneous binding of man6P and IGF-II.
A soluble, truncated form (~240 kDa) representing the extracellular domain circulates in serum and retains binding capability for both ligands.
