The monoclonal antibody 27D6 recognizes the CD85d (ILT4/LIR-2) protein, a member of the ILT/LIR/MIR receptor family. CD85d is a 110 kDa transmembrane receptor with three Ig-like domains and a cytoplasmic tail containing ITIMs (immunoreceptor tyrosine-based inhibitory motifs). Expression is confined to monocytes, macrophages, and dendritic cells.

CD85d binds non-classical class I HLA-G, -B, and G1 molecules. Upon ligand binding, tyrosine phosphatase SHP-1 interacts with ITIMs, contributing to immune tolerance during fetal implantation and potentially influencing organ allograft rejection.
Antibody 27D6 has been shown to block ligand (HLA-G tetramer) binding.

LILRB2 (ILT4) gene belongs to the leukocyte immunoglobulin-like receptor (LIR) family located on chromosome 19q13.4. The encoded receptor contains multiple extracellular Ig-like domains, a transmembrane region, and cytoplasmic ITIM motifs.
It is expressed on immune cells where it binds MHC class I molecules on antigen-presenting cells, transmitting inhibitory signals that regulate immune activation, inflammation, and cytotoxicity. Multiple transcript variants encoding different isoforms have been identified.
