Product Specific Information

The MEM-166 monoclonal antibody reacts with human CD177, a 58–64 kDa GPI-anchored glycoprotein. The human CD177 locus encodes at least two alleles, NB1 and PRV-1, differing by four amino acids. CD177 is expressed on granulocytes, early erythroblasts, megakaryocytes, promyelocytes, and myelocytes. CD177 mRNA levels are elevated in several myeloproliferative disorders such as polycythemia vera. Antibodies to CD177 are associated with neonatal alloimmune neutropenia, autoimmune drug-induced neutropenia, and marrow graft failure.

Applications Reported:
This MEM-166 (MEM166) antibody has been reported for use in flow cytometry, immunoprecipitation, and Western blotting (non-reducing conditions).

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood. Recommended ≤1 µg/test (100 µL final volume). Cell counts may vary (10⁵–10⁸ cells/test). Optimal dilution should be determined empirically.

Target Information

CD177 (NB1/HNA-2a and PRV-1 form) is a GPI-anchored glycoprotein primarily expressed on neutrophils. Its expression increases during pregnancy, inflammation, or after G-CSF application. The ligand of CD177 is CD31 (PECAM-1). CD177 is involved in neutrophil transmigration and may act as a pro-proliferative molecule. Antibodies against CD177 are associated with neonatal alloimmune neutropenia (NAN).
