Product Specific Information

The TW7-16B4 monoclonal antibody reacts with mouse latency associated peptide (LAP, pro-TGF beta 1, LAP/TGF beta 1). TGF beta mediates effects on proliferation, differentiation, and function of many cell types. It is synthesized as a precursor containing LAP at the N-terminus and mature TGF beta at the C-terminus. Cleavage between amino acids 278 and 279 forms LAP and TGF beta dimers, which associate to form the latent TGF beta complex. LAP is secreted and found in the extracellular matrix, and can also be expressed on platelets and activated regulatory T cells via binding to GARP (LRRC32).

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on stimulated mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number: 10^5–10^8 cells/test. Titrate for optimal performance.

PerCP-eFluor™ 710 can replace PE-Cy5, PE-Cy5.5, or PerCP-Cy5.5. Excitation at 488 nm, emission at 710 nm. Recommended filters: 685 LP dichroic mirror and 710/40 band pass (695/40 acceptable alternative).

PerCP-eFluor™ 710 conjugated antibodies remain stable after overnight exposure to freshly prepared 2% formaldehyde at 4°C.

Target Information

TGF beta-1 is a cytokine found ubiquitously in tissues, stored in the extracellular matrix as a latent complex with its prodomain. Activation requires aV integrin binding to an RGD sequence and mechanical force, mediated by latent TGF-beta binding proteins (LTBP). Latent forms are complexes of TGF-beta, TGF-LAP (latency associated peptide), and LTBP.
