Product Specific Information
The V3GYWCH monoclonal antibody reacts with human and mouse interferon regulatory factor 8 (IRF8; ICSBP). IRF8 is a 50 kDa transcription factor essential for the development and differentiation of dendritic cells (DC), particularly plasmacytoid DC (pDC) and CD8a+ DC. IRF8 deficiency leads to impaired DC maturation and function.
Applications Reported:
Used for intracellular staining followed by flow cytometric analysis.
Applications Tested:
Validated by intracellular staining and flow cytometry of normal human peripheral blood cells using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended usage ≤ 0.5 µg/test (100 µL reaction volume). Optimal titration advised.
Fluorochrome Characteristics:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable).
Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.

Filtration:
0.2 µm post-manufacturing filtered.
Target Information
IRF8 (Interferon Regulatory Factor 8) acts as a transcriptional activator or repressor. It binds upstream regulatory regions of type I IFN and IFN-inducible MHC class I genes. IRF8 negatively regulates immune cell function and participates in CD8+ dendritic cell differentiation via interaction with BATF-JUNB complex, activating immune-specific genes. Mutations may cause immunodeficiency types 32A and 32B.
