Takara pRI 909 and pRI 910 Binary Vectors
The Ri pRI 909 and pRI 910 binary vectors are designed for Agrobacterium-mediated plant transformation and contain a T-DNA region suitable for transformation of Arabidopsis, tomato, tobacco, and rice.
The T-DNA originates from the vir region-lacking Rhizobium (Agrobacterium) rhizogenes Ri plasmid.
Both pRI 909 and pRI 910 function as shuttle vectors, capable of autonomous replication in E. coli and Rhizobium (Agrobacterium).
In E. coli, these vectors achieve a high copy number due to the presence of the ColE1 replication origin.
Both ColE1 ori and the mutant-type Ri plasmid replication origin (Ri-ori) are stably maintained in Rhizobium (Agrobacterium).
The vectors carry the kanamycin-resistance gene NPTIII for selection in E. coli and Rhizobium (Agrobacterium), and a mutant-type kanamycin-resistance gene NPTII for selection in plants.
The pRI 909 and pRI 910 vectors enable efficient plant transformation via a binary vector system with Rhizobium (Agrobacterium).
Stable integration of target genes into plant chromosomes is facilitated by cloning sites located near the T-DNA Right Border (RB), minimizing the risk of deletion of the target gene.
