Product Specific Information

The SK3 monoclonal antibody reacts with human CD4, a 59-kDa cell surface receptor expressed on most thymocytes, a subset of mature T helper cells, and at lower levels on monocytes. CD4 serves as a receptor for HIV, and SK3 can block HIV binding and mixed lymphocyte reactions. SK3 and RPA-T4 antibodies recognize distinct epitopes.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells. Recommended usage: 5 µL (0.1 µg) per 100 µL test volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before mixing with cells.
  • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology, offering engineered fluorescent signatures with optimized spillover and spread characteristics.

Target Information

CD4 is a co-receptor for MHC class II recognition and HIV binding. It is expressed in T helper lymphocytes, monocytes, macrophages, dendritic cells, and in tissues such as thymus, lymph nodes, tonsils, spleen, and certain brain and gut regions. CD4 participates in T-cell activation via association with the T-cell receptor complex and Lck kinase. Multiple alternatively spliced transcripts exist (RefSeq, July 2017).
