Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in the cytoplasm and cell membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19.

CD22 membrane expression is limited to mature B cells (CD22+) and absent in plasma cells (CD22−), making it useful for phenotyping mature leukemias. It is strongly expressed in hairy cell leukemia and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.

CD22 preferentially binds α2,6-linked sialic acid, and its recognition site can be masked by cis interactions. Upon ligand-induced tyrosine phosphorylation, CD22 regulates B-cell receptor signaling, interacting with Src family kinases and recruiting phosphatases that modulate signal transduction.
