Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (low-affinity Fc gamma RIII). CD16 has two isoforms, FcγRIIIA and FcγRIIIB.

• FcγRIIIA is expressed on monocytes, macrophages, and NK cells.
• FcγRIIIB is detected on neutrophils as a GPI-anchored form.

Expression patterns are similar in non-human primates, though species differences exist (e.g., neutrophil expression in sooty mangabeys).
Binding of IgG triggers antibody-dependent cellular cytotoxicity (ADCC), phagocytosis, cytokine release, and antigen presentation.

Epitope information: 3G8 recognizes the same epitope as CB16 but distinct from B73.1.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 4 µL (0.6 µg)/test (final volume 100 µL, 10⁵–10⁸ cells/test).

NovaFluor dye compatibility:

• Not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

Staining recommendation:
Add 5 µL of CellBlox Blocking Buffer per sample (or per 100 µL of cell suspension for single-color controls).

Target Information

CD16 (FCGR3A) encodes a receptor for the Fc portion of IgG, mediating immune complex clearance and cellular cytotoxicity.

• FCGR3A: Expressed on NK cells, membrane-anchored.
• FCGR3B: Expressed on neutrophils, PI-anchored.
  Mutations in FCGR3A are associated with Immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and neonatal neutropenia.
  Alternative splicing yields multiple isoforms.
