Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16, also known as the low-affinity Fc gamma RIII.
CD16 exists as two isoforms, Fc gamma RIIIA and Fc gamma RIIIB.

• Fc gamma RIIIA: expressed on monocytes, macrophages, and NK cells.
• Fc gamma RIIIB: expressed on neutrophils as a GPI-anchored form.

Expression patterns vary among primate species. Binding of IgG triggers signal transduction leading to ADCC, phagocytosis, cytokine release, and antigen presentation.

3G8 recognizes the same epitope as CB16 but binds distinct epitopes compared to B73.1.
Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells.
Use at 5 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) required for optimal staining.
Add 5 µL per sample before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor Technology:
Based on Phiton technology using nucleic acid dye structures engineered for minimal spillover and spread.

Target Information

CD16 encodes an Fc receptor (FCGR3A) that binds immunoglobulin G and mediates immune complex clearance, ADCC, and viral infection enhancement.

• FCGR3A: expressed on NK cells (transmembrane anchored).
• FCGR3B: expressed on neutrophils (PI linkage).

Mutations are associated with Immunodeficiency 20, Herpes Zoster, and systemic lupus erythematosus susceptibility.
