Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16, also known as low-affinity Fc gamma RIII.
CD16 exists as two isoforms: FcγRIIIA (on monocytes, macrophages, NK cells) and FcγRIIIB (on neutrophils).
3G8 recognizes the same epitope as CB16 but distinct from B73.1.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells
  • Use 4 µL (0.6 µg) per test
  • A test = antibody amount staining 100 µL cell sample (10⁵–10⁸ cells/test)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Add 5 µL of CellBlox Blocking Buffer per sample when staining with NovaFluor conjugates.
• For single-color controls: use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD16 encodes a receptor recognizing the Fc portion of IgG, involved in immune complex clearance and cell-mediated cytotoxicity.
Two genes, FCGR3A and FCGR3B, encode CD16 isoforms:

• FCGR3A: expressed on NK cells, membrane-anchored
• FCGR3B: expressed on neutrophils, PI-anchored

Mutations are associated with immunodeficiency 20, recurrent viral infections, systemic lupus erythematosus, and neonatal neutropenia.
Alternative splicing produces multiple isoforms.
