Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16 (Fc gamma RIII).
CD16 exists as two isoforms, FcγRIIIA and FcγRIIIB.

• FcγRIIIA: Expressed on monocytes, macrophages, and NK cells.
• FcγRIIIB: Found on neutrophils (GPI-anchored form).

Expression patterns differ among primates: in macaques and baboons, CD16 is not found on neutrophils, whereas in sooty mangabeys it is present.
CD16 binding to IgG triggers ADCC, phagocytosis, cytokine release, and antigen presentation.

• Clone 3G8 recognizes the same epitope as CB16, but distinct from B73.1.
• Each product includes 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.
• Reported application: Flow cytometric analysis.
• Tested use: 5 µL (0.6 µg) per 100 µL test volume (10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before adding to cells.
  • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton nucleic acid dye structures, engineered for optimized fluorescence with minimal spillover and spread.

CD16 encodes an Fc receptor (FcγRIII) that binds the Fc portion of IgG, mediating immune complex clearance and antibody-dependent cellular cytotoxicity.

  • FCGR3A – expressed on NK cells (transmembrane form)
  • FCGR3B – expressed on neutrophils (PI-linked form)
• Associated Diseases: Immunodeficiency 20, Herpes Zoster, systemic lupus erythematosus susceptibility, and alloimmune neonatal neutropenia.
