Target Information
The RANKL (TNFSF11/CD254) gene encodes a member of the TNF cytokine family. It acts as a ligand for osteoprotegerin and plays a key role in osteoclast differentiation and activation. It also functions as a dendritic cell survival factor and regulates T cell–dependent immune responses.
Activation of T cells induces expression of this gene, promoting osteoclastogenesis and bone loss. RANKL activates antiapoptotic kinase AKT/PKB through a signaling complex involving SRC kinase and TRAF6, suggesting a role in apoptosis regulation.
Disruption of the related gene in mice leads to severe osteopetrosis, absence of osteoclasts, defects in T and B lymphocyte differentiation, and failure to form mammary structures during pregnancy. Two alternatively spliced transcript variants have been identified.
