Product Specific Information

This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by subsets of tissue macrophages such as splenic red pulp macrophages, Kupffer cells, and intestinal lamina propria macrophages. Mouse monocytes do not express CD163, and it is not a reliable M2 macrophage marker in mice.

This antibody detects CD163 on fixed and permeabilized cells, enabling intracellular staining. Aggressive tissue dissociation may reduce surface CD163; intracellular detection is recommended.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤1.0 µg/test (100 µL final volume).

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is suitable for cytometers with a 355 nm UV laser. Protect from light due to photo-oxidation sensitivity.

Fixation and Storage:
Samples may be fixed in IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in the dark. Compatible with short-term methanol-based fixation (≤1 hour).

Recommended Buffers:
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) when combining multiple polymer dye-conjugated antibodies to reduce non-specific interactions.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It serves as a receptor for the hemoglobin-haptoglobin complex, protecting tissues from oxidative damage and aiding iron recycling via endocytosis.

CD163 is expressed on human monocytes and macrophages, upregulated by IL-10, IL-6, and dexamethasone, and shed upon stimulation with LPS or PMA. It binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner and triggers intracellular signaling leading to IL-6 and CSF1 secretion.
