Product Specific Information

The 14D3 monoclonal antibody reacts with human CD152 (CTLA-4), a protein structurally related to CD28 and expressed on activated T cells. CTLA-4 binds CD80 (B7-1) and CD86 (B7-2) with higher affinity than CD28, delivering inhibitory signals that limit T cell activation and IL-2 secretion.

The 14D3 clone also recognizes rhesus monkey CTLA-4 and exhibits inhibitory activity.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of stimulated human peripheral blood cells using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).

For complete detection of CTLA-4, both surface and intracellular expression should be evaluated due to its intracellular localization.

Fluorochrome Information:
Brilliant Ultra Violet™ 805 (BUV805) emits at 797 nm and is optimized for cytometers equipped with a 355 nm UV laser. Protect from light due to photo-induced oxidation sensitivity.

Staining Recommendations:
When using multiple Brilliant or polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) or Brilliant Stain Buffer (Product # 00-4409-75) to minimize non-specific interactions.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark. BUV805 is compatible with short-term methanol fixation (<1 hour).

Target Information

CTLA4 (Cytotoxic T-Lymphocyte Antigen 4) is a CD28-family receptor expressed mainly on CD4+ T cells. It binds CD80 and CD86 with higher affinity than CD28, delivering inhibitory signals that downregulate T cell activation. Intracellular CTLA4 is also expressed in regulatory T cells and contributes to their suppressive function.
