Product Specific Information

Description:
The monoclonal antibody 3E4 reacts with human and mouse interferon regulatory factor 4 (IRF4), a 52 kDa transcription factor involved in B cell, T cell, and macrophage function.

  • In B cells, IRF4 is highly expressed in mature plasma cells and regulates differentiation via interaction with PU.1 and transcription of B cell–specific genes such as Prdm1 (Blimp1).
  • In T cells, IRF4 contributes to Treg, Th2, Th9, and Th17 cell development and induces Blimp1 and IL-10 expression.
  • IRF4 is also involved in macrophage polarization and regulation.
  • Overexpression of IRF4 has been observed in various hematologic malignancies.

Applications Reported:
For intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested using intracellular staining and flow cytometric analysis of stimulated normal human peripheral blood cells with the Foxp3/Transcription Factor Buffer Set (Product #00-5523-00).
Recommended use: ≤ 0.25 µg per test (100 µL final volume; 10⁵–10⁸ cells/test).
Antibody should be titrated for optimal performance.

Light Sensitivity:
PE-Cyanine7 is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

Excitation/Emission: 488–561 nm / 775 nm
Laser Compatibility: Blue, Green, Yellow-Green Laser
Filtration: 0.2 µm post-manufacturing filtered


Target Information

The IRF (interferon regulatory factor) family of transcription factors features a unique tryptophan pentad repeat DNA-binding domain. IRFs regulate interferon responses to viral infection and control interferon-inducible genes.
IRF4 is lymphocyte-specific and negatively regulates TLR signaling, a key pathway in innate and adaptive immunity.
A chromosomal translocation t(6;14)(p25;q32) involving IRF4 and IgH may contribute to multiple myeloma.
Alternative splicing variants have been identified.
