Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human LIF. Anti-Human LIF-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA:
To detect Human LIF by sandwich ELISA (using 100 µL/well), a concentration of 0.25–1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human LIF (500-P39) as a capture antibody, allows the detection of at least 0.2–0.4 ng/well of Recombinant Human LIF.

Western Blot:
To detect Human LIF by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible development reagents, the detection limit for Recombinant Human LIF is 1.5–3.0 ng/lane under either reducing or non-reducing conditions.

Target Information

Leukemia inhibitory factor (LIF) is a 20 kDa protein that belongs to the IL-6 receptor family. It binds to a heterodimeric membrane receptor composed of a LIF-specific subunit (gp190 or LIFR) and gp130, shared with other IL-6 family members. LIF is expressed in tissues such as thymus, lung, and neuronal tissue, and can be upregulated by cytokines like TNFα and IL-17. Elevated LIF levels are associated with rheumatoid arthritis, neural injury, systemic inflammation, and tuberculosis. LIF inhibits differentiation of embryonic stem cells in mice and supports stem cell self-renewal. Human and mouse LIF share 79% sequence homology and exhibit cross-species activity, though stem cell differentiation inhibition is mouse-specific.
