Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human MIP-1β (CCL4). The specific antibody was purified by affinity chromatography and biotinylated.

Sandwich ELISA:
To detect Human MIP-1β by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human MIP-1β (500-P38B) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human MIP-1β.

Western Blot:
For detection of Human MIP-1β by Western Blot, use at 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human MIP-1β is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

CCL4 (macrophage inflammatory protein 1-beta, MIP-1β) belongs to the intercrine beta (chemokine CC) family. It plays roles in chemotaxis, proinflammatory response, and immune homeostasis. Produced by macrophages upon stimulation by bacterial endotoxins, CCL4 recruits and activates various inflammatory cells. It is expressed by lymphocytes, macrophages, and dendritic cells.
CCL4 and CCL3 regulate trafficking and activation of inflammatory cells during infection by bacteria, viruses, parasites, and fungi. While both affect monocytes similarly, CCL4 selectively attracts CD4+ lymphocytes, whereas CCL3 attracts CD8+ lymphocytes. Both are potent chemoattractants for B cells, eosinophils, and dendritic cells.
Processed CCL4 can down-modulate CCR5 receptor expression, inhibiting CCR5-mediated HIV-1 entry in T cells. CCL4 binds with high affinity to CCR5 receptors.
