Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human LIF. Anti-Human LIF-specific antibody was purified by affinity chromatography employing an immobilized Human LIF matrix.
Sandwich ELISA:
To detect Human LIF by sandwich ELISA (using 100 µL/well antibody solution), a concentration of 0.5–2.0 µg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human LIF (500-P39Bt) as a detection antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human LIF.
Western Blot:
To detect Human LIF by Western Blot analysis, this antibody can be used at a concentration of 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human LIF is 1.5–3.0 ng/lane, under either reducing or non-reducing conditions.
Target Information
Leukemia inhibitory factor (LIF) is a 20 kDa protein belonging to the IL-6 receptor family. It binds to a heterodimeric receptor composed of a LIF-specific subunit (gp190 or LIFR) and the shared subunit gp130. LIF expression is found in thymus, lung, and neuronal tissues. It can be up-regulated by pro-inflammatory cytokines such as TNFα and IL-17. Elevated LIF levels have been associated with rheumatoid arthritis, neural injury, systemic inflammation, and tuberculosis. LIF is known for inhibiting differentiation of embryonic stem cells in mice, contributing to stem cell self-renewal. Human and mouse LIF share 79% sequence homology and exhibit cross-species activity, though stem cell differentiation inhibition is mouse-specific.
