Target Information
The Ragulator complex consists of three proteins—C11orf59, ROBLD3, and MAPKSP1—which target mTORC1 to lysosomal surfaces. This complex plays a role in endosomal trafficking and can be exploited by viruses such as coronavirus to enhance replication.
C11orf59 is regulated by cholesterol levels, participating in LDL uptake and cholesterol homeostasis. It interacts with PPAR-alpha to control lipid and carbohydrate metabolism in diabetes.
Most effects of C11orf59 are associated with mTORC1 kinase-mediated growth promotion, and dysregulation of the mTOR pathway may have metabolic implications.
