Product Specific Information
Reconstitution:
Briefly centrifuge the vial before opening. Reconstitute the lyophilized protein in water to a concentration of 0.1–1.0 mg/mL. Do not vortex.
Store the reconstituted solution at 2–8°C for up to 1 week. For long-term storage, dilute in a buffer containing a carrier protein (e.g., 0.1% BSA) and store aliquots at -20°C to -80°C.Storage:
The lyophilized protein is stable at room temperature for 1 month, but should be stored at -20°C to -80°C for long-term stability, preferably desiccated.
Working aliquots stored with carrier protein are stable for at least 12 months at -20°C to -80°C. Avoid repeated freeze/thaw cycles.
Target Information
TGF beta-1 (TGFB1, Transforming Growth Factor Beta 1) is a polypeptide member of the transforming growth factor beta superfamily of cytokines, found almost ubiquitously in tissues. It is stored in the extracellular matrix as a latent complex with its pro-domain. Activation requires the binding of αV integrin to an RGD sequence in the prodomain, exerting force on this domain, which is held in the extracellular matrix by latent TGF-β binding proteins (LTBP).
Latent forms are complexes of TGF beta-1, an amino-terminal portion of the TGF-beta precursor (TGF-LAP, latency-associated peptide), and a specific binding protein (LTBP). TGF beta-1 regulates proliferation, differentiation, adhesion, and migration in many cell types. Many cells have TGF beta receptors, and the protein both positively and negatively regulates other growth factors.
The mature peptide may form heterodimers with other TGF beta family members. The gene for TGF beta-1 is frequently upregulated in tumor cells, and mutations in this gene are associated with Camurati-Engelmann disease and cystic fibrosis.
