Product Specific Information

  • For immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein of the TNF receptor superfamily. It was originally identified as a cell surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1. The ligand for CD30 is CD30L (CD153), and their interaction mediates effects such as cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a crucial role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas. It acts as a costimulatory molecule in thymic negative selection and is expressed on Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (including Burkitt’s), virus-infected T and B cells, and activated normal T and B cells. In T cells, CD30 is expressed on subsets producing Th2-type cytokines and on CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL4 receptor.

The soluble form (sCD30) serves as a marker reflecting Th2 immune response. CD30 interacts with TRAF2 and TRAF5, leading to NF-κB activation, and functions as a positive regulator of apoptosis. It limits the proliferative potential of autoreactive CD8 effector T cells, contributing to protection against autoimmunity. Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
