Product Specific Information

  • IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was initially identified as a surface antigen of Hodgkin and Reed-Sternberg cells using monoclonal antibody Ki-1.
CD30 binds to its ligand CD30L (CD153), mediating diverse cellular responses such as proliferation, activation, differentiation, and apoptosis. It plays a crucial role in the pathophysiology of Hodgkin’s disease and other CD30-positive lymphomas and acts as a costimulatory molecule in thymic negative selection.
Expression of CD30 is observed in Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin lymphomas (e.g., Burkitt’s lymphoma), virus-infected T/B cells, and activated normal T/B cells. In T cells, CD30 is expressed on subsets producing Th2 cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4R.
The soluble form (sCD30) serves as a marker for Th2 immune responses. CD30 interacts with TRAF2 and TRAF5, leading to NF-κB activation. It functions as a positive regulator of apoptosis and limits autoreactive CD8 effector T cell proliferation, contributing to immune regulation.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
