Product Specific Information

  • For immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
  • Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was originally identified as a surface antigen of Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1.
CD30 binds to its ligand CD30L (CD153), mediating diverse cellular effects including proliferation, activation, differentiation, and apoptosis. It plays a critical role in the pathophysiology of Hodgkin’s disease and other CD30⁺ lymphomas, and acts as a costimulatory molecule in thymic negative selection.
CD30 expression is found in Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (e.g., Burkitt’s lymphoma), virus-infected T/B cells, and activated normal T/B cells. In T cells, CD30 is expressed on Th2-type cytokine-producing subsets and CD4⁺/CD8⁺ thymocytes co-expressing CD45RO and IL4 receptor.
The soluble form (sCD30) serves as a marker of Th2 immune response. CD30 interacts with TRAF2 and TRAF5, leading to NF-κB activation. It positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, contributing to protection against autoimmunity.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
