For immunohistochemistry (PFA-fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily. It was first identified as a surface antigen on Hodgkin’s and Reed-Sternberg cells using monoclonal antibody Ki-1.
CD30 interacts with its ligand CD30L (CD153), mediating various effects including cell proliferation, activation, differentiation, and apoptosis. It plays a key role in Hodgkin’s disease and other CD30+ lymphomas and acts as a costimulatory molecule in thymic negative selection.
CD30 is expressed in Hodgkin’s and Reed-Sternberg cells, certain non-Hodgkin’s lymphomas (e.g., Burkitt’s lymphoma), virus-infected T and B cells, and activated normal T and B cells. In T cells, CD30 is found on subsets producing Th2-type cytokines and on CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor.
Soluble CD30 (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30, mediating NF-κB activation. CD30 positively regulates apoptosis and limits autoreactive CD8 effector T cell proliferation, protecting against autoimmunity.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported. CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, most Anaplastic Large Cell Lymphomas (ALCL), and embryonal carcinomas.
