Product Specific Information
The 2H7 monoclonal antibody reacts with human CD20, a 33–36 kDa transmembrane protein expressed on developing and mature B cells, but not on plasma cells. CD20 may play a role in B-cell activation and is occasionally found at low levels on a subset of mature T cells.
Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Fluorochrome Information:
eFluor® 450 is an alternative to Pacific Blue®, excited at 405 nm and emitting at 445 nm. Ensure the instrument supports this fluorochrome.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, as well as a subset of T lymphocytes and follicular dendritic cells. It regulates calcium conductance, cell cycle progression, and B-cell proliferation. CD20 is associated with lipid rafts and colocalizes with BCR during activation. It remains on the cell surface after antibody ligation, making it a valuable target for therapeutic B-cell depletion. CD20 dysfunction is linked to Ms4a1-related common variable immune deficiency.
