열매개 항원 회수(heat-mediated antigen retrieval)는 염색 전 권장됩니다.
10mM citrate buffer (pH 6.0)에서 10분 처리 후 실온에서 20분 냉각합니다.
항원 회수 후, 1차 항체를 실온에서 30분간 반응시킵니다.
양성 대조 조직으로는 대장암(colon carcinoma)을 권장합니다.

Lewis 혈액형 시스템은 외분비 상피세포에서 합성되어 체액 내에 순환하는 푸코실화된 글리코스핑고지질 세트로 구성됩니다.
이 글리코스핑고지질은 발생, 조직 분화, 종양 전이, 염증, 세균 부착에 관여하며, 적혈구에 흡착되어 Lewis 표현형을 형성합니다.
Lewis 항원 생합성의 마지막 단계에서 푸코스를 전구 다당류에 첨가하는 효소를 암호화하는 푸코실트랜스퍼라아제(fucosyltransferase) 계열 유전자에 의해 조절됩니다.
이 유전자의 변이는 대부분의 Lewis 항원 음성 표현형의 원인입니다.
대체적으로 동일한 단백질을 암호화하는 여러 가지 스플라이스 변형체가 존재합니다.
