Product Specific Information

This M4G3LN monoclonal antibody recognizes human and mouse Bruton’s tyrosine kinase (BTK) and interleukin-2-inducible tyrosine kinase (ITK) when phosphorylated on Y551 and Y511, respectively.
Tec family kinases, including BTK and ITK, are critical for activation of phospholipase C and calcium-dependent signaling following antigen receptor triggering. BTK is involved in B cell signaling via TLR4 co-receptor RP105/CD180. Deficiency or mutation in BTK or ITK leads to defects in B or T cell development and signaling, such as X-linked agammaglobulinemia.

Specificity of the M4G3LN clone was confirmed by ELISA, flow cytometry, and western blotting.

Applications Tested:

• Immunoblotting of stimulated Daudi B cells: ≤5 µg/mL
• IHC (formalin-fixed paraffin-embedded human tissue, high pH antigen retrieval): ≤0.1 µg/mL
  It is recommended to titrate the antibody for optimal performance.

Target Information

Tec family kinases (including BTK and ITK) are receptor-proximal kinases expressed in B and T cells.
BTK mediates B cell activation, while ITK functions in T cell signaling.
Mutations or deletions in BTK result in X-linked agammaglobulinemia, and ITK deficiencies impair T cell signaling.
