Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events such as the establishment and maintenance of pregnancy.

PR exists as two forms:

  • PR-A (94 kDa): Contains a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses transcriptional activity.
  • PR-B (120 kDa): Transcriptionally active form responsible for gene activation related to endometrial maintenance, pregnancy, and ovulation inhibition.

In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Binding of progesterone induces a conformational change, enabling receptor dimerization and binding to progesterone response elements (PRE), leading to activated transcription.

A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
