Product Specific Information

The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes through surface CD14.

Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells
• Recommended use: 5 µL (1.0 µg) per test in 100 µL final volume, with 10⁵–10⁸ cells/test

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01)

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample to antibody cocktails before combining with cells
• For single-color controls: use 5 µL per 100 µL of cell sample (10³–10⁸ cells)

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 binds lipopolysaccharide (LPS) via LBP, contributing to immune recognition and inflammatory response. It also binds apoptotic cells and plays roles in allergic, inflammatory, and infectious processes.
Associated diseases include Mycobacterium chelonae infection and croup.
