Product Specific Information

The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes through surface CD14.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Recommended usage: 5 µL (1.0 µg) per test in 100 µL final volume. Cell number: 10⁵–10⁸ cells/test.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide (LPS) receptor and is released into serum as both secreted and cleaved forms. CD14 binds LPS molecules via LBP, aiding in immune recognition and modulation of inflammatory responses. Soluble CD14 (sCD14) can distinguish structural differences in LPS and plays roles in neutralization, apoptosis recognition, and immune signaling. CD14 dysfunction is associated with infections such as Mycobacterium chelonae and Croup.
