Target Information

Keyhole limpet hemocyanin (KLH) is a large, heterogeneous glycosylated protein composed of subunits (350,000–390,000 Da) forming aggregates of 4,500,000–13,000,000 Da. Each domain contains two copper atoms that bind a single oxygen molecule (O₂), giving the molecule a transparent blue color when oxygenated.

KLH is highly immunogenic and safe for human use, making it valuable as a vaccine carrier protein. Due to its complexity, KLH cannot be synthesized artificially and is purified from the marine mollusk Megathura crenulata.

KLH is widely used as a carrier protein for haptens (low molecular weight substances such as peptides, small proteins, and drug molecules) to elicit an immune response. Its large size, multiple epitopes, and abundance of lysine residues enable high hapten-to-carrier ratios, enhancing hapten-specific antibody production. Being phylogenetically distant from mammals, KLH minimizes cross-reactivity in mammalian research systems. However, it can aggregate, which, while still immunogenic, may complicate conjugation and laboratory handling.
