Target Information
CXCR4 (also known as Fusin, LESTR, HUMSTR) is a G-protein-coupled chemokine receptor with seven membrane-spanning domains. It functions as a co-receptor for X4 HIV-1 entry into CD4+ cells and also mediates HIV-2 binding and fusion.
CXCR4 is the receptor for the chemokine CXCL12/SDF-1, which triggers intracellular calcium increase and MAPK1/MAPK3 activation. It also acts as a receptor for extracellular ubiquitin, influencing calcium and cAMP levels.
The receptor plays critical roles in hematopoiesis, cardiac and vascular development, and neuronal survival. CXCR4 binds bacterial LPS and mediates inflammatory responses, including TNF secretion by monocytes. Antibodies to CXCR4 can block HIV-1 and HIV-2 fusion and infection. The amino-terminal domain and second extracellular loop serve as HIV binding sites.
